373 Abnormal type VII collagen expression in non-lesional psoriatic skin

The healthy-looking non-lesional (NL) psoriatic skin already carries some alterations. These alterations include extracellular matrix defects such as the disruption of laminin networks, which may lead to impaired keratinocyte adhesion basal membrane. This process is associated with aberrant expression α5β1 integrin complex and extradomain-A-containing fibronectin (EDA+FN) around keratinocytes. In our previous work, in NL EDA+FN was increased compared healthy (H) skin. Fibronectin can specifically bind type VII collagen therefore, we aimed examine presence function H We found thatthe displayed reduced by immunostaining transmission electron microscopy. Type protein decreased western blot. detected TH domain polyclonal antibody but not monomeric form collagen. Cultured fibroblast from showed while there no change observed keratinocytesby RT-qPCR pSTAT3 activation Blocking STAT3 resulted fibroblasts it didn’t derived fibroblasts, indicating role regulation human fibroblasts. results provide additional evidence about altered tissue organization
